Summary of the proposal : Defects in genes encoding for proteins involved in mitochondrial beta oxidation of fatty acids result in diverse metabolic disorders, with broad clinical phenotypes from very severe forms with neonatal death, to non-life threatening mild forms of disease. For most of these disorders there is a poor biochemical and structural understanding of the consequences of a particular mutation, and the available biomarkers are restricted to acyl-carnitine profiles which are determined by MS/MS as part of newborn screening programs. Typical therapeutic strategies rely on oral intake of riboflavin (vitamin B2) which is sometimes combined with coenzyme Q, whose modes of action are not however fully understood. In addition, there is growing evidence that other cellular processes are also affected, as a result of a defect in a beta-oxidation enzyme. In this context, the field would benefit from a broader analysis of the metabolites accumulating under disease conditions.

With this proposal we seek to combine our expertise in order to i) characterise new biomarkers for diagnosing FAO defects; ii) understand the effect of current drugs at the metabolic level. This will be achieved by: a) using NMR to analyse the metabolic profiles corresponding to mitochondria from patient fibroblasts and b) implementing Drosophila as a disease model.
