Stabilome E coli
Perspectives of extension of this common project to a more ambitious proposal to be submitted to funding agencies : Long term Added value of the cooperation : The labs have different expertises required for the project : the stabilome determination and analysis for LISBP and RNAses activities and regulation for ITBQ. The association of the two competences will be pioneer in the field, leading to interesting joint poublications.
Summary of the proposal :
This exchange is planned in the frame of a project in collaboration between the two laboratories and both teams are highly interested. The labs have different expertises and the results will be pioneer in the field, leading to interesting joint poublications.
This project aims at identifying the mRNA targets of two main exoribonucleases in E. coli, PNPase and RNaseR, through genome-wide stability analysis using microarray technology (=”stabilome”).
It is now well known that gene expression involves several regulatory mechanisms at various level. Among those, mRNA degradation is critical and is the focus of this project.
The experiments will be perfomed using 3 differents strains: a wild type MG1655 that will be the reference strain, a PNPase deletion mutant and a RNaseR deletion mutant. Both mutants will be constructed in ITQB. The studies will be performed in stationnary phase of growth during which PNPase as well as RNaseR levels are known to be increased. After the first growth assays, Muriel Cocaign-Bousquet will come to ITQB for a project meeting. At this time the time of rifampicin addition for the transcription arrest (the exact OD or time after entry into stationary phase) will be defined.
During the 3 months she will spend in LISBP, Clémentine Dressaire will perform the stabilome experiments using the total RNA samples that she would have extracted in ITQB before in the determined conditions. Data analysis will also be performed there with the support of statistics specialits. Before the end of these three months Cécilia Arraiano will visit for a project meeting.
